- Autoradiograph of in vitro RCNMV replicase products. Partially purified RCNMV replicase
fraction was assayed as described by Hayes and Buck (1990). Ten microcuries of 32P-a-ATP was
added to a final reaction volume of 25 ul. No exogenous RNA templates were added. The replication
products were electrophoretically separated on a native TAE agarose gel (1.2%). Replicase products
were detected by autoradiography. Two replicase products with mobility close to 4 kb and 1.6 kb DNA
standard were detected from the S100 fraction (lane 1) and F3 fraction (lane 3) purified from the infected
tissues. Fractions S100 (lane 2) and F3 (lane 4) purifed from healthy tissue did not produce any
radioactive products.
